As editor of the two-part quantity V on photosynthesis in RUHLAND'S Encyclopedia, the forerunner of this sequence released in 1960, i've been approached via the editors of the current quantity to supply a quick preface. The justification for following this advice lies within the nice alterations that have been occurring in biology within the 20 years among those courses, alterations that are mirrored within the new editorial plan. 20 years in the past it seemed handy and officially effortless to contemplate picture synthesis as a essentially separated box of study, that could be handled lower than significant headings: one featuring essentially photochemical and biochemical prin ciples, the opposite physiological and environmental experiences. this type of partition, in spite of the fact that, so far as goals and evaluations of the authors have been involved, ended in a slightly heterogeneous quantity. this present day, the tendency in experimental biology is in the direction of a merger of formerly unique disciplines. Biochemists and biophysicists have constructed their how to such an volume that, over and above the research of person response sequences, paintings at the manifold interrelationships between mobile actions has develop into in creasingly attainable. becoming a member of them in becoming numbers are the physiologists and ecologists with their wealth of knowledge on job adjustments in vivo and at the variability and potency of the organisms involved. moreover, biochemists, biophysicists and physiologists additionally now percentage a full of life curiosity in ultrastructure study, the implications and implications of which, via consistently enhancing method, have generated very important stimuli for the paintings within the box of phone function.

Other investigations of direct photoassimilation of phosphorus by intact cells gave results that were at best suggestive (see review, ARNON, 1956). In short, experiments with whole cells proved, for reasons discussed below, incapable of yielding evidence for photoreduction of pyridine nucleotides and for a special, light-induced phosphorylation. The occurrence of these light reactions in photosynthesis was discovered not in whole cells but in isolated chloroplasts. 1. ARNON: chemically generated from their respective precursors within the chloroplasts.

Because the protein under its various names was isolated from, and effective in, chloroplasts, it was thought at first to be a unique chloroplast protein. However, in 1961 the association of the protein with chloroplasts ceased to be unique when K. TAGAWA and M. NOZAKI (unpublished data from this laboratory) and LOSADA et al. (1961) isolated a protein with similar properties from the photosynthetic bacterium Chromatium. Although Chromatium cells do not have chloroplasts, do not photoreduce NADP+, and do not evolve oxygen, the "pyridine nucleotide reductase" isolated from these bacterial cells was able to replace the native protein of spinach chloroplasts in mediating the photoreduction of NADP+ and the concomitant evolution of oxygen.
